全球十大最常见导致死亡的疾病当中唯一一种缺乏有效治疗方案的疾病是什么？ 是阿尔茨海默病。所谓的有效的标准就是如果有药物或其他方法能改善阿尔茨海默病患者的症状。这期聊聊阿尔茨海默病治疗的新进展。 认知衰退是一个全球性的健康难题。世界上每 3 秒就会有一人患上认知衰退。2020 年，全世界有超过 5500 万人患有阿尔茨海默病。这一数字几乎每 20 年翻一番，到 2030 年将达到 7800 万人，到 2050 年将达到 1.39 亿人。老年人口增长最快的国家是中国、印度及其南亚和西太平洋邻国。 阿尔茨海默病不仅致命，而且在发病后的数年或数十年间，这种疾病会逐渐剥夺患者的记忆力、思维能力和生活自理能力，最终甚至会让患者失去基本的人权和尊严。如果你看过2014年拍的电影《永远的爱丽丝》Still Alice 的话，里面讲述了一个语言学教授Alice的患病经历，真是让人心碎。 阿尔茨海默病为何难治？ 阿尔茨海默病之所以难治是因为它的致病成因复杂。阿尔茨海默病的病原并不是大脑的不正常运行，像癌细胞的不停繁衍，或是像心脏病患者中动脉粥样硬化导致血管堵塞那样。阿尔茨海默病根源在于大脑巨大的神经突触网络系统的自然缩减。但这个缩减的速度太快了。也就是说，在阿尔茨海默病患者当中是大脑的正常运行程序失控了。 阿尔茨海默病的三大类型 阿尔茨海默病实际上是大脑对某些威胁的保护性反应。问题在于，当这些威胁持续、反复且强烈发生时，大脑的反应也会持续、反复且强烈，最终会诱使这些保护性措施越界而异化成为有害性的。阿茨海默病往往是大脑针对代谢异常毒素等危害因素的过度保护后引发的。 Apollo Health 的首席科学官, 著名的神经学家 Dale Bredesen

癌症被誉为“众病之王”，全球每年有数百万人饱受各种类型的癌症之苦。以世界卫生组织公布的2023年的数据为例。按发病率计算，中国十大癌症与全球相比略有不同，其中肺癌、结直肠癌和甲状腺癌占据前三大癌症类型。 全球新晋药王是什么 2023年一款叫做 Keytruda（可瑞达）或者帕博利珠单抗的药物以250亿美元的销售额，成为全球新晋药王。Keytruda 由制药公司默克 （Merck & Co.）,（在美国和加拿大以外称为MSD（Merck Sharp & Dohme））默沙东开发。 Keytruda 是一种免疫疗法药物，在 2014 年首次获得美国食品药品管理局 (FDA) 批准，用于治疗晚期黑色素瘤，此后又被批准用于治疗多种其他类型的癌症。 这期聊聊目前癌症治疗领域最火热的研究领域–免疫疗法。 免疫疗法治疗思路 免疫疗法是一种很有前景的抗癌新方法。现在常用的癌症治疗方法像化疗和靶向治疗都是把肿瘤当成敌人。免疫治疗则是开启了一个新的时代。人体每七分钟就会产生一个癌细胞。之所以大多数人都没有得癌症，是免疫环境帮我们清除了它们。免疫疗法的理念就是调动人体自身的力量来调整免疫状态，使我们更好地对抗癌症。它的原理不是针对敌人的弱点，而是致力于调整自身的土壤，让机体回归正常。 CAR-T 细胞疗法 免疫疗法的治疗思路之一是CAR-T细胞疗法。它通过利用患者自己的免疫系统来对抗癌细胞。 采集T细胞：首先，从患者的血液中提取T细胞，这是一种白细胞，通常在人体内起到抵抗感染和疾病的作用。 改造T细胞：接下来，科学家在实验室中对这些T细胞进行基因改造。通过加入一种称为嵌合抗原受体（CAR）的特殊受体，这些T细胞能够更有效地识别和攻击癌细胞。 扩增T细胞：经过改造后，科学家在实验室中大量培育这些改造后的CAR-T细胞，以确保有足够数量来对抗癌症。 回输T细胞：最后，这些改造后的CAR-T细胞被输回患者体内。它们会在体内寻找并攻击癌细胞。 CAR-T 细胞疗法的优势在于它可以靶向特定的癌细胞。比如，Keytruda 通过阻断一种免疫细胞

2024年上汽集团宣布其与固态电池生产商清陶能源共同研发的第一代固态电池已在排产宣布，旗下的智己，飞凡等等车型都会搭载固态电池，实现量产。2025年将会达到十万辆级规模的目标。 7月11号，大众汽车集团旗下的电池公司 PowerCo 和美国的固态锂电池生产商 QuantumScape (NYSE: QS) 宣布，双方已达成一项开创性协议，将 QuantumScape 的下一代固态锂金属电池技术工业化。 这期我们来聊聊固态电池的新进展。我用上市公司美国的 QuantumScape 和 2024 胡润全球独角兽中国的清陶能源举例。 数据显示，在一辆电动汽车中, 电池会占到整车成本的30%~40%。而动力电池铁三角–快充，续航能力和安全性，一直是新能源汽车消费者最关心的问题。 目前市面上用的锂离子电池，在正负极之间有一种液态电解质，这样锂离子在电池充电时就能朝一个方向流动，而在电池耗电时则朝另一个方向流动。传统的锂离子电池正在逼近容量上的物理极限，所以全球很多厂商都在专注研发固态电池。固态电池顾名思义，就是用固体材料代替液体，其中使用的固体材料包括陶瓷和硫化物等等。 固态电池并不新鲜，迈克尔 法拉第早在 19 世纪就发明了固态电解质。但是之所以不能量产，QuantumScape 的联合创始人 Jagdeep Singh 总结了四个方面的原因： 它们需要高电流密度，这样才能快速充电。如今的固态设计只能在低电流密度下工作， 充电速度不够快。 内置的锂不能过多。过多的锂会导致储存的能量减少。 需要持久耐用。有些固态设计在充放电几百次后就会失效。这些电池需要经过数十万英里的行驶才能发挥商业价值。
